Further information: | The plasmid was constructed by replacing the VF1 fragment in p416-VF1 by the fragment encoding amino acids 1-172 of yEVenus, which was amplified from pKT103, using the SpeI/XbaI and BspEI restriction sites. This plasmid is meant for multicolor bimolecular fluorescence complementation in yeast, which allows the in vivo determination of multiple protein interactions at a time. The plasmid encodes a yeast codon-optimised enhanced version of Venus (yEVenus), which is the fastest maturing yellow variant of the A. victoria GFP protein. The N-terminal fragment of the yEVenus coding sequence is followed by a multiple cloning site. Other names of the plasmid are pNS1499 and p416-YFP-N for N-term tagging. |
